No, it appears to be separated into packs.

However if you buy both then:

As a bonus for purchasing both packs - as a bundle or separately - you can get eight different-coloured Yoshis and eight different-coloured Shy Guys that can be used right away.

Red/Blue/Green Gold Peach confirmed.

Although I like any additional content regardless (excluding skins in most cases) I can't help but feel that there are so many missed opportunities here. First off nobody wanted another Mario or Peach clone (though somebody probably wanted Dry Bowser) and the fact that Wario's Gold Mine is here means that there's no chance of Wario's Colosseum returning as well.

But I am looking forward to the new tracks and the retro tracks that do make it in even if they aren't the ones I want, I don't mind the crossover thing at all, I still hope that the final new track is themed after Super Mario Galaxy rather than like Star Fox, Metroid or even Sonic Colors or something.

I originally thought that guest characters from other Nintendo franchises should have debuted in the next Mario Kart, but I've changed my mind. I'll pre-order the MOMENT this becomes official.

I'm not even going to preorder it because it is DLC. Anyone who can afford DLC can get it. There is absolutely no reason why they can't just sell DLC on the E-Shop to unlimited amounts of people.
