Heck, even WarioWare got a course before Wario Land. (but then again Namco developed that game and they're more willing than Nintendo to put references from other Mario games such as characters in their Mario games for some reason)

Captain Syrup is a horrible addition. She just wouldn't fit. I'd rather see Baby Wario get added before that.

I would rather have Captain Syrup than Baby Wario. We need another female character that's not royalty, and Captain Syrup plays important roles in the game she appears in and she's a freaking female pirate.

If they want another female, I'd rather see Dixie Kong, and heck, even Tiny Kong before those.

Judging with recent game appearance, the girls I think should be in are Pauline, Dixie and Mona from Wario games.
I don't hate any of them, and it's unreasonable to say any of them shouldn't join Mario Kart. They felt that Waluigi was basically made as a counterpart to Luigi like Wario is to Mario, they wanted to have Peach have a Wario-esque counterpart, but Miyamoto refused to allow it to happen. Check Mario Tennis 64 page on the wiki and look in the trivia.
What's funny is that I found out about this today before that comment was made. Ironic?

Oh God. Thanks Miyamoto for not letting that happen. Can you imagine? We would have had that thing in almost every single Mario spin-off starting with Mario Party 3. Then again, I would like to have a main female villain introduced in the series.

Captain Syrup already made her debut before Tennis 64. Coincidentally Camelot also features human athletes. If "counterparts" just means cloning princess and plumbers, both Syrup and Nina/Kate from MarioTennis GBC are much more creative than W-Peach obviously.
From the Camelot history, it seems that Nintendo and Miyamoto had argued for Golf 64 due to the overuse of generic human character, which is lack of Marioverse characteristics. However, both Golf 64 and Tennis 64/Open are two different extremes and lack of balance.
Back to topic, besides Syrup who is clearly a boss type, many other characters can be Peach's rival for different reason (Pauline: imaginary rival in love, Mona: cooperated with Wario). Hope Nintendo can consider to give Peach a rivalry story in spinoffs by putting more playable girls.

Never really liked Coin Runners. I think Bob-Omb battle needs to return. (Not the "only bombs" option, I am talking about the one in Double Dash)
Also I'd like to see Mission Mode like in DS, although fleshed out into a full story mode.

Yeah, why not? Mission mode should definitely return IMO. I think coin runners, baloon fight, and shine theif would all be good too. Throw in mirror mode time trials and I'll be happy with just about any roster

Birdo has appeared in quite a few spin-offs so there's no reason to off her now. Personal opinion makes a dream roster. Odds and reasoning make a believable one. That's why I said those changes are irrelevant to your roster.
